Is there any way to restrict users in PPTP VPN to just single logins and not multiple logins from different locations?

You should be able to accomplish this by editing the profile assigned to the user. In Winbox under the user profile click limits tab then "Only One" check Yes .

Terminal ; ppp profile set only-one=yes default
change default to the profile name.
