Vlan on one port

I have never had contact with the VLAN.
I have:
1 rb450g
1 Unmanaged Switch, 48-port
25 desktops computer
25 laptops

I would like to separate the laptop from the desktop computer.
Is it possible to connect one single cable from the switch to rb450g and create two VLANs?

Yes, but then you’re relying on all your client devices to tag their traffic [=either impossible or a nightmare to manage]. You really need a managed switch. Another possibility would be mixing two L3 networks on one L2 network but then at least half of your clients wouldn’t be able to use DHCP.
