Is it possible to have multiple Hotspot on one hardware interface (eg. eth3)?
I am trying to achieve multiple hotspot for different locations connected through VPN to the HQ - Main router, wich will host the Hotspot?

One hotspot per logical interface. So you can use VLANs to get multiple hotspots on one physical interface for example. I'm not sure if you can run a hotspot service over an EoIP tunnel, but you could try that.
