connect 2 SFP+ switches

Hi all
Please could you help me in this case ?
I have 2 switches a TP Link with 4 SFp+ port and Mikrotik CRS 309 with 8xSFP+ ports when i have all server connected to the Mikrotik Switch alle is running fine see Picture this is the switch below in the Picture Also the Internet is working what is connected to the switch on the top of the picture in one RJ45 Port.

But when i connect as example one server to to the other switch "Red Line" then i cant reach this server --Ping is working sucessfull -- webinterface on the port 5000 in the browser has network error.
Both Switches running in default settings
What is wrong or what is missing?

I have a few questions,

On the top switch, the far left RJ45 and far right SFP+, are those different VLAN's? Why do you have both plugged in? If they are separate VLAN's, why not just send both VLAN's down the SFP+ port and do away with that RJ45 connection?

That top switch, the red line, are you sending multiple VLAN's down that port? Is that a tagged or untagged port?
